Sudan IV is more soluble in fat than in alcohol allowing fat to absorb the stain.

RECOMMENDED PROCEDURE: NOTE: Keep Sol. I tightly stoppered since the evaporation of the alcohol and acetone causes precipitation of the stain. Dip for an instant in Sol. II. Stain in Sol. I for 2 - 5 minutes. Wash quickly in Sol. II and transfer to distilled water. Counterstain in Sol. III. Wash thoroughly in water and mount in glycerin.
RESULTS: Nuclei - blue; fat - orange to red; normal myelin and fatty acids - unstained.
