FOR IN VITRO DIAGNOSTIC USE ONLY.
RECOMMENDED PROCEDURE: Specimens - intestinal (calciform cells), tracheal and embryonic tissue. Stain in Sol. I for 5 - 10 minutes. Rinse in 95% Ethanol. Transfer to Sol. II until the slide appears dark green. Dehydrate, clear and mount in balsam. Mix two parts of Solution IA - Methylene Blue with one part Solution IB - Crystal Violet. Place stain mixture onto slide for 10 - 15 seconds. Afterwards, allow the excess stain to run off the slide. Add Bismarck Brown for 45 seconds. Rinse slide with tap water. Allow slide to dry and then view.
RESULTS: Cartilage - dark brown; mucin - light brown; nuclei of all the cells - green. Examine under oil immersion at 1000X with direct illumination. Blue violet is positive (either entire cell or intracellular granules); yellow - brown is negative.
