Technique used in the staining of distemper inclusion bodies.
CONTENTS: Hematoxylin, Potassium Alum, Ethanol, HCl, Biebrich Scarlet, Orange G, Fast Green FCF, Phosphotungstic Acid, Phosphomolybdic Acid, Glacial Acetic Acid
These solutions are made from certified dyes (when applicable).
FOR IN VITRO DIAGNOSTIC USE ONLY.
RECOMMENDED PROCEDURE: Sol. I - 3 - 5 minutes. Rinse in water. Differentiate in Sol. II until cytoplasm is free of hematoxylin (check microscopically). Blue in ammonia water. Wash in running water -10 minutes. Sol. III - 1 minute. Rinse in 95% ethanol (check microscopically for clear, light-green connective tissue.) Dehydrate in 95% ethanol, absolute ethanol, and clear in xylene, 2 changes each.
RESULTS: Inclusion bodies - brilliant red; connective tissue - light green; elastic tissue - purplish red; muscle - red; keratin - orange; erythrocytes - orange red; nuclei - blue.
