RECOMMENDED PROCEDURE: Working Solution: In the following order, mix 20 ml of Sol. I + 8 ml of Sol. II + 8 ml of Sol. III.
NOTE: This staining solution should,be used within 24 hours. Immerse sections in working solution for 15 minutes to 1 hour until perfectly black. Differentiate in Sol. IV for a few minutes. Examine sections in water under low magnification to observe the stages in differentiation. If the differentiation has been carried too far, (provided they have not been treated with alcohol) the sections may be restained. Wash in water. Place in 95% alcohol to remove iodine and leave sections in tap water 5 minutes or longer. Counterstain 3 - 4 minutes in Sol. V. Clear in xylene and mount.
RESULTS: Elastic fibers - intensely blue-black; nuclei - blue to black; collagen - red.
