RECOMMENDED PROCEDURE: Bacteria: Fixative - absolute alcohol or Carnoy’s fluid. Stain 2 - 4 minutes (up to 30 minutes if necessary). Wash in distilled water. Dehydrate quickly in absolute alcohol or acetone. Clear in xylol and mount in balsam. Celloidin sections are cleared in terpineol and mounted in balsam.
RESULTS: Bacteria - bright red; chromatin - green; all other structures stain varying shades of red.
RECOMMENDED PROCEDURE: DNA and RNA: Fixative - Clarke and Camoy’s fluid. Stain 10 minutes. Rinse twice in distilled water briefly. Blot with smooth filter paper.
Differentiation Solution: Mix 30 ml Tertiary Butyl Alcohol with 10 ml 100% Alcohol. Dehydrate in a fresh change of the differentiating solution for 1 - 2 minutes. (Slides may be left in solution for 4 - 5 minutes) Clear in two changes of xylene for 10 minutes each.
RESULTS: DNA - blue green; RNA - bright red; remainder of tissue - pale pink.
RECOMMENDED PROCEDURE: Plasma cells: Fixative - absolute alcohol or Camoy’s fluid. Stain 20 minutes. Rinse rapidly in water. Differentiate and dehydrate rapidly in absolute alcohol. Clear in xylol.
RESULTS: Nuclei - green; cytoplasm - deep red.
