An excellent nuclear stain employing a half-oxidized hematoxylin solution. (Gill, 1974) Gill Hematoxylin is a high contrast stain and a progressive hematoxylin which enchances the use of counterstains. To achieve desired stain characteristics in different specimens, the stain is available in 3 concentrations at 0.2% w/v, (Modified), 0.4% w/v, and 0.6% w/v. The modified solution at 0.2% may be preferred in cytology, however, by increasing the time (10 minutes) it can be used in histology. Gill-Hematoxylin at 0.4% is recommended for histology, however, a shorter stain time renders this solution suitable for cytologic use. For comparable results to those obtained with Harris’ Hematoxylin, Gill-Hematoxylin at 0.6% is recommended. An essential part of this stain sequence is the use of a blueing agent. We recommend Scott’s Tap Water Substitute which ensures a controlled pH and aids in cell retention on slides.

RECOMMENDED PROCEDURE: Gill-Hematoxylin (Modified): Fix in Sol. III for 15 minutes. Water - 2 x (10 dips each). Gill-Hematoxylin (Modified) - 2 minutes. Water - 2 x (10 dips each). Sol. II - 1 minute. Water - 2 x (10 dips each). Sol. III - 2 x (10 dips each). Continue with preferred staining method. Gill-Hematoxylin 0.4 % or 0.6 %: Xylene - 3 x (2 minutes each). Absolute Ethanol - 3 x (30 seconds each). Water - 2 x (10 dips each). Gill Hematoxylin 0.4% or 0.6% - 3 minutes. Water - 2 x (10 dips each). Sol. II - 2 minutes. Water - 2 x (10 dips each). Continue with preferred staining method.

RESULTS: Nuclear chromatin stains blue to blue-black; cytoplasm faintly tinted.
